Association of CYP1B1 Polymorphisms with Breast Cancer: A Case-Control Study in the Han Population in Ningxia Hui Autonomous Region, P. R. China

چکیده
Studies investigating possible associations between cytochrome P4501B1 (CYP1B1) polymorphisms and breast cancer risk have been inconsistent. We set out to ascertain whether there might be an association between polymorphisms in exon 2 (codon 119, G-->T) and exon 3 (codon 432, G-->C) of CYP1B1 and breast cancer in a Chinese Han population in the rural region of Ningxia. Using an allele-specific polymerase chain reaction method and direct DNA sequencing, the presence or absence of the two CYP1B1 polymorphisms was investigated. Genotype and allele frequencies were analyzed in breast cancer cases (n = 152) and healthy age-matched controls (n = 156). The odds ratio (OR) of 119G-->T or 432G-->C in breast cancer cases and controls was 3.3 (95% CI: 1.28 to 8.28) and 2.8 (95% CI: 1.04 to 7.51), respectively. In addition, the OR for people with both polymorphisms (119T and 432C) was 4.69 (95% CI: 1.97 to 11.19). Our results suggest that certain polymorphisms in the CYP1B1 gene might increase risk for breast cancer among Han Chinese, perhaps because they influence the efficiency of CYP1B1 bio-transformation of oestrogens or pro-carcinogens into DNA-reactive electrophiles that may act as cancer-initiating agents.

No association between single nucleotide polymorphisms in pre-mirnas and the risk of gastric cancer in Chinese population
Objective(s): Accumulating evidence has demonstrated that miRNAs contribute to various genetic and epigenetic modifications in the pathogenesis of gastric cancer (GC). Recent studies focused on the four single nucleotide polymorphisms (SNPs) of pre-miRNAs including rs11614913, rs3746444, rs2910164, and rs2292832. It was suggested that these four SNPs were significantly associated with the risk ...
متن کاملComparison of Genetic Variants in Cancer-Related Genes between Chinese Hui and Han Populations
BACKGROUND The Chinese Hui population, as the second largest minority ethnic group in China, may have a different genetic background from Han people because of its unique demographic history. In this study, we aimed to identify genetic differences between Han and Hui Chinese from the Ningxia region of China by comparing eighteen single nucleotide polymorphisms in cancer-related genes. METHODS...
متن کاملAssociation of Obesity Related Genetic Variants (FTO and MC4R) with Breast Cancer Risk:a population-based case-control study in Iran
Background: The heterogeneous breast cancer is the most common cause of cancer-related mortality. Obesity defined by BMI is known as a major risk factor for breast cancer. Objective: The purpose of this study was to explore the role of obesity related-polymorphisms rs9939609 FTO and rs17782313 MC4R in breast cancer development. Materials and Methods: We obtained matched peripheral blood, serum ...
متن کاملGenetic polymorphisms in the estrogen receptor - α Gene codon 325(CCC}CCG) and risk of breast cancer among Iranian women: a case control study
Abstract Background: The Iranian breast cancer patients are relatively younger than their Western counterparts. Evidence suggests that alterations in estrogen signaling pathways , including estrogen receptor-α (ER- α ), occur during breast cancer development in Caucasians. Epidemiologic studies have revealed that age-incidence patterns of breast cancer in Asians differ from those in Cauca...
متن کاملThe Association of the MTHFR Gene Polymorphisms with Breast Cancer Susceptibility
Introduction: Breast cancer is the most common malignancy in women worldwide. It is also the second leading cause of cancer death among women after lung cancer. Considering the relationship among plasma folate levels, the level of uracil, and DNA damage in cell division, methyl tetrahydrofolate reductase (MTHFR) is a suitable candidate for studies on the susceptibility to cancer, including brea...
